Is it possible to upload a kml/kmz file to define site names and physical locations? We would like to have locations pre-defined in the map-messages options so our users can click on a location.

We don't support a direct import of kml / kmz files but I'd like hear more about what you are trying to do. We're always looking for ways to simplify and streamline the management of organizational data like this.
In the meantime, I think the best way to automate the management of sites is to use our REST API.
There is a POST Site web service to create and update sites here.
There is a GET Site web service to query sites here.
